ABSTRACT

Damage by soil borne plant pathogens is a major threat in crop production and there are increasing concerns on biological control. A trial was made to suppress bacterial wilt of tomato (BWT) caused by Ralstonia solanacearum using biocontrol agents (BCAs) isolated from soils in Kalimantan, Indonesia. A crystal violet medium was used to isolate Gram-negative bacteria. A total of 270 isolates of gram negative bacteria were obtained from forest and agricultural peat soils and sandy soils. Five isolates were selected as better performing BCAs to BWT through four times screenings using a pumice medium. The isolates were nearest to Burkholderia nodosa, B. pyrrocinia, B. sacchari and B. terricola, according to the 16S rDNA sequences, and fatty acid composition. All of these BCAs had phosphate solubilizing ability and survived well in acidic condition.

ABSTRAK

Salah satu faktor dominan yang menyebabkan kerusakan tanaman adalah patogen tular tanah sehingga perlu pengendalian alternatif menggunakan agensia hayati. Penelitian telah dilakukan untuk mengendalikan penyakit layu bakteri tomat yang disebabkan oleh Ralstonia solanacearum menggunakan agensia hayati yang diisolasi dari tanah Kalimantan (daerah hutan dan pertanian gambut serta tanah berpasir), Indonesia. Media kristal violet digunakan untuk mengisolasi bakteri gram negatif sejumlah 270 isolat. Lima isolat terbaik terpilih setelah melalui empat kali skrining pada media pumice. Isolat yang ditemukan tersebut adalah Burkholderia nodosa, B. pyrrocinia, B. sacchari and B. terricola berdasarkan 16S rDNA sequencing dan pengujian komposisi asam lemak menggunakan FID-GC/MS. Agensia hayati ini mempunyai kemampuan untuk melarutkan fosfat dan survive pada kondisi asam.
